Chinese hamster ovary cell line EM-C11 was obtained by ethylnitrosourea induced mutagenesis of CHO-9 fibroblast cells (GM16132). It is sensitive to simple alkylating agents and has a 10 fold elevated level of sister chromatid exchanges. EM-C11 is a member of x-ray cross complementation group XRCC1. It is defective in the repair of DNA single strand breaks and in DNA base excision repair. EM-C11 is defective in the XRCC1 gene. In the base excision repair mechanism the product of the XRCC1 gene forms a complex with DNA ligase III. |
