Hereford breed. The age of the donor is estimated. The thoracic aorta was removed immediately post-mortem on 10/22/85. The culture was initiated from cells released from the lumen of the vessel by collagenase digestion. The cell morphology is polygonal. The culture tested positive for endothelial cell-specific von Willebrand factor and negative for alpha-smooth muscle actin (immunofluorescence assays). The karyotype is 60,XY; normal diploid male. The first and second passage cells were propagated in medium supplemented with horse plasma-derived serum and heparin (0.1mg/ml) on gelatin-coated flasks. The culture was grown on gelatin-coated flasks and the medium was supplemented with ECGS (0.02 mg/ml) and heparin (0.05 mg/ml). Culture medium: AMMC 15U with supplements. For lifespan determination this cell line was cultured in Ham's F12 medium with 10% inactivated fetal bovine serum. This cell line will only be shipped frozen.
