About this Collection

The Allen Cell Collection is a bank of human induced pluripotent stem cells that are currently derived from the WTC parental line released by the Conklin Laboratory at the J. David Gladstone Institute (available through Coriell as GM25256, where you can also find the de-identified patient information associated with the line). Please refer to the Allen Institute for Cell Science publication for more details on how these cells lines were generated along with the extensive quality control data (Roberts et al., MBoC 2017).

To answer the most basic questions about human health and disease, we need to understand how the dynamic organization and activities of the cell drive its function. This set of powerful gene edited human stem cells from the Allen Institute for Cell Science is the first publicly available collection of hiPSC lines with key structural proteins fluorescently tagged. All gene editing and quality control were performed by the Allen Institute for Cell Science.

Cell Catalog Update

Three new fluorescently tagged hiPSC lines are now available in the Allen Cell Collection. The first line is a dual edited cell line labeling mitochondria (via Tom20) and microtubules (via alpha-tubulin). The other two are single edited, nuclear cell lines in which DNA replication foci (via PCNA) or CTCF-binding sites (via CTCF) are visualized.

The Allen Cell Collection now has 51 high quality-certified hiPSC lines available to help your research program. The focus of the Collection is fluorescently-tagged hiPSC lines that target 40 key cellular structures/substructures. The Allen Cell Collection is now available to academic researchers and commercial entities! New lines continue to be released several times a year, so stay tuned for new cell lines.

Please pay special attention when ordering either of the endosome or Golgi lines as the AICS number is now qualified with a three-digit clonal extension to help discriminate between the mono- and bi-allelic edits of the same structure.

Newest lines available in 2021:

Nuclear:

TERF2-mEGFP-mono

PNCA-mEGFP-mono

CTCF-mEGFP-mono

Dual line:

TOMM20-mEGFP-mono TUBA1B-mTagRFP-T-mono

Quality Assurance

Each cell line has undergone a broad range of quality control experiments to ensure precise insertion of the fluorescent tag and localization of the tagged protein to the correct cellular compartment without disrupting the cells’ overall genomic integrity. More details about how these lines are generated and the Quality Control steps involved in their generation are available by clicking on the AICS link provided for each line in the “Order Page” for each cell line.
