Induced Pluripotent Stem Cells

Coriell’s iPSC Core offers a variety of services to meet your stem cell needs. Coriell’s iPSC Laboratory has extensive expertise establishing hiPSC lines, reprogramming various ‘parental’ cell types using different methods including Sendai viral and episomal plasmid vectors. Coriell has a large and growing collection of apparently healthy and disease model iPSC lines. Disease specific cell lines include neurological disorders, monogenic disorders, and chromosomal abnormalities.

Over the decades, Coriell has played a pivotal role in helping to define and implement best practices for biospecimen collection, processing (ensuring sterility, integrity and authenticity), quality assurance (QA), quality control (QC), cryopreservation, sample safe storage, collection maintenance/inventory management, and global distribution. Coriell’s laboratories operate under the most stringent standard operating procedures following ISO:9001 standards and cell line resources undergo comprehensive cell line integrity and QC testing.

In the case of stem cell lines, assessment of pluripotency and differentiation potential is performed using a variety of technological platforms. Standard cell line QC assays performed at Coriell (and offered as a service to the scientific community) include, but are not limited to:

  • Growth and viability assessment
  • Microbial and fungal sterility testing
  • Mycoplasma testing
  • Human pathogen testing
  • Identity testing (microsatellite matches with the parent cell culture)
  • Cytogenetic G-banded karyotype
  • Microarray genotyping
  • Confirmation of integration-free reprogramming
  • Assessment using stem cell surface markers via immunocytochemistry and/or flow cytometry and alkaline phosphatase staining
  • In vitro embryoid body (EB) assay and quantitative analysis of tri-lineage differentiation potential using RT-PCR to analyze expression of germ layer specific genes 
  • Pluripotency and tri-lineage differentiation potential using Applied Biosystems TaqMan hPSC Scorecard Panel.
